摘要:

研究芽孢杆菌属菌株Bacillus velezensis JS8的次生代谢产物。采用硅胶柱色谱和制备高效液相色谱技术从B. velezensis JS8的ISP2液体培养基发酵物中分离纯化出4个化合物,并通过质谱和核磁共振技术进行结构鉴定,分别为desmethyl venezueline G(1)、N-(5-formyl-2-hydroxyphenyl)acetamide(2)、N-(2-hydroxy-5-methylphenyl)acetamide(3)、3-acetamido-4-hydroxybenzoic acid(4),其中化合物1为新化合物。通过HPLC分析不同发酵天数发酵液的代谢成分,推测化合物1~4可能的生物合成代谢途径。并对分离得到的化合物进行抗菌活性测试(金黄色葡萄球菌与大肠杆菌),结果显示,化合物3对金黄色葡萄球菌具有一定的抗菌活性,其最小抑菌浓度为18.1 μg/mL。此外,对所有化合物进行宫颈癌HeLa细胞毒活性以及脂多糖诱导的RAW 264.7细胞释放NO的抑制活性测试,化合物1~4在50 μmol/L浓度下均无明显的细胞毒活性及NO释放量抑制活性。

Abstract:

This study aims to investigate the secondary metabolites produced by the Bacillus velezensis JS8.Four compounds were isolated from ISP2 liquid fermentation products of the B. velezensis JS8 by silica gel column chromatography and preparative high performance liquid chromatograph,and their structures were identified by the NMR and MS data as a new metabolite desmethyl venezuel G (1) and three known metabolites N-(5-formyl-2-hydroxyphenyl)acetamide (2),N-(2-hydroxy-5-methylphenyl)acetamide (3),and 3-acetamido-4-hydroxybenzoic acid (4) Based on the HPLC analysis of culture broths in different culture days,the biosynthetic pathway of compounds 1-4 was proposed.Compounds 1-4 were tested for antibacterial activities against Staphylococcus aureus and Escherichia coli.The results showed that only compound 3 had moderate antibacterial activity against S.aureus with minimum inhibitory concentration (MIC) value of 18.1 μg/mL.In addition,all compounds were evaluated for cytotoxicity against HeLa cell line and anti-inflammatory activity via inhibiting NO production in lipopolysaccharide-stimulated RAW 264.7 cells in vitro.The results indicated that compounds 1-4 showed no cytotoxic activity and no inhibitory effect against NO production with concentration of 50 μmol/L.
