基于UPLC-Q-TOF-MS/MS分析蒙药狼毒毒效部位的化学成分

Components analysis of the toxic and effective parts of the Mongolian medicine Euphorbiae Fischerianae Radix by UPLC-Q-TOF-MS/MS

摘要:

利用超高效液相色谱-四极杆飞行时间质谱联用技术(ultra performance liquid chromatography quadrupole time of flight mass spectrometry,UPLC-Q-TOF-MS/MS)分析蒙药狼毒(狼毒大戟)毒效部位化学成分,比较狼毒石油醚、二氯甲烷、乙酸乙酯、正丁醇部位所含化合物的差异。结果表明,通过文献及实验室建立的数据库对比,解析得68个化合物,其中有43个二萜类、5个三萜类、8个香豆素类、2个酚酸类、2个鞣酸类、3个苯乙酮类、1个甾醇类、1个生物碱类、1个脂肪酸类、1个间苯三酚类和1个其他类化合物。本研究采用的UPLC-Q-TOF-MS/MS技术为鉴定狼毒毒效部位化学成分提供了快速、高效的定性分析方法。该研究不仅为狼毒毒效部位化学成分研究以及狼毒毒性评估提供了科学依据,也为狼毒药用价值的进一步开发提供了实验基础。

Abstract:

The chemical components of the toxic and effective parts of the Mongolian medicine Euphorbiae Fischerianae Radix were analyzed using ultra performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-Q-TOF-MS/MS).The differences in the compounds contained in the petroleum ether,dichloromethane,ethyl acetate,and n-butanol fractions of Euphorbiae Fischerianae Radix were compared.The results showed that,through comparison with the literature and the database established in the laboratory,68 compounds were identified.Among them,there were 43 diterpenoids,five triterpenoids,eight coumarins,two phenolic acids,two tannins,three acetophenones,one sterol,one alkaloid,one fatty acid,one phloroglucinol,and one other type of compound.The UPLC-Q-TOF-MS/MS technique used in this study provides a rapid and efficient qualitative analysis method for identifying the chemical components of the toxic and effective parts of Euphorbiae Fischerianae Radix.This study not only offers a scientific basis for the research on the chemical components of the toxic and effective parts of Euphorbiae Fischerianae Radix and the toxicity assessment of this herb,but also provides experimental data for the further development of the medicinal value of Euphorbiae Fischerianae Radix.
