云木香根茎中的倍半萜类成分研究

摘要:

采用大孔树脂、正相硅胶、反相硅胶等色谱层析材料及半制备高效液相色谱等色谱层析技术,从云南丽江产云木香根茎中分离鉴定了9个倍半萜类化合物,包括愈创木烷型3个(化合物1、2和9)、桉叶烷型3个(化合物4~6)和吉玛烷型3个(化合物3、7和8)。运用波谱光谱学方法分别鉴定为:去氢木香内酯(1)、sausinlactone A(2)、木香烃内脂(3)、α-环木香烃内酯(4)、β-木香酸(5)、reynosin(6)、picriside B(7)、saussureamines A(8)和B(9)。其中,化合物8和9为较少见的倍半萜和氨基酸的杂合体。采用MTT法测试了化合物1、3、4、6、8和9对肝癌细胞株(HepG2)的体外细胞毒活性,结果表明这些化合物在30 μM浓度下对HepG2 肝癌细胞株显示弱的抑制活性。其中,化合物3对肝癌细胞株(HepG2)的抑制率为60%。

Abstract:

Nine sesquiterpenoids,including three guaiane-type (1,2 and 9),three eudesmane-type (4-6) and three germacrane-type (3,7 and 8),were isolated from MeOH extract of the rootstock of Saussurea lappa C.B Clarke by silica gel and Rp18 column chromatography,as well as semi-preparative high performance liquid chromatograph.The compounds were identified as dehydrocostus lactone (1),sausinlactone A (2),costunolide (3),α-cyclocostunolide (4),β-costic acid (5),reynosin (6),picriside B (7),saussureamines A (8) and B (9) by NMR and Mass spectroscopy.Among them,compounds 8 and 9 were unnormal aminoacid-sesquiterpene lactone conjugates.The MTT method was used to test cytotoxic activity of compounds 1,3,4,6,8 and 9 against liver cancer cell lines (HepG2).These compounds showed weak inhibit activity against HepG2 liver cancer cell lines at a concentration of 30 μM.Among them,the inhibit activity of compound 3 was more than 60%.
