Purification process of total flavonoids from Eucommia ulmoides leaves based on Box-Behnken response surface method

Abstract:

This study investigated the separation and purification of total flavonoids from Eucommia ulmoides leaves using macroporous resins.Static and dynamic adsorption tests were used for resin screening,and single-factor experiments were performed to analyze parameter effects on purification efficiency.A comprehensive index considering adsorption,desorption,and recovery rates was adopted for evaluation,and response surface experiments were designed to optimize process conditions.In the 12 tested resins,AB-8 resin showed the best performance for flavonoid adsorption and separation.The optimal parameters were confirmed as:sample loading concentration 2.78 mg/mL,loading volume 3.55 BV,impurity removal with 4 BV ultrapure water,elution with 75% ethanol at a flow rate of 2 BV/h.Under these conditions,the comprehensive score reached 73.60%,and the purity of total flavonoids was improved 4.11 times.This simple,practical optimized process provides a basis for further research on Eucommia ulmoides leaves flavonoids.
