Comparison of Three Chromatographic Methods for Monosaccharide Composition Analysis of Tremella Polysaccharide

Abstract: To find an accurate and fast method to determine the monosaccharide composition of Tremella polysaccharide, thin layer chromatography (TLC), gas chromatography (GC) and high performance liquid chromatography (HPLC) were compared. The results of TLC and GC were not ideal. However, HPLC was a simple method with high sensitivity, good resolution and integrity information. The result of HPLC analysis showed that the monosaccharide compositions were Glc, Man, GlcA, Xyl and Fuc, with the mole percentage of 0.24:1.00:0.06:0.29:0.25. Consequently, HPLC is the most suitable method for monosaccharide composition analysis.
