补肾活血方中单体成分的体外炎性抑制作用研究

In vitro Anti-Inflammatory Activity of Effective Components in Bushen Huoxue Recipe

摘要: 本文采用LC-MS技术进行定性分析,确定补肾活血方水提液中存在芍药苷、阿魏酸、白藜芦醇、蛇床子素及补骨脂素等单体化合物。利用酶联免疫法(ELISA)测定试药对模型细胞释放TNF-α、IL-6及NO的抑制作用。与空白组比较,模型组TNF-α、IL-6、NO含量明显升高(P<0.05);与模型组比较,阿魏酸、白藜芦醇、补骨脂素及复方水提部位在100 μg/mL,以及蛇床子素在20、100 μg/mL均对TNF-α的释放具有显著的抑制(P<0.01);阿魏酸及补骨脂素在100 μg/mL,白藜芦醇及蛇床子素在20、100 μg/mL均对IL-6水平具有显著的抑制(P<0.01);芍药苷及复方水提部位在100 μg/mL,阿魏酸、白藜芦醇、蛇床子素及补骨脂素在20、100 μg/mL均对NO的释放具有显著抑制(P<0.01)。结果表明单体对炎性因子的释放抑制作用总体上强于复方水提部位,提示补肾活血方的治疗骨关节炎活性可能是通过抑制炎症介质释放,降低TNF-α、IL-6、NO等含量发挥作用,这对验证复方药效活性,探究效应物质基础具有重要意义。

Abstract: Qualitative analysis of paeoniflorin,ferulic acid,resveratrol,osthole and psoralen in water extract of Bushen Huoxue recipe was performed using LC-MS technology.Enzyme-linked immunosorbent assay (ELISA) was applied to detect the effect of paeoniflorin,ferulic acid,resveratrol,osthole,psoralen and Bushen Huoxue Recipe water extract on IL-6,TNF-α and NO.The results showed that ferulic acid (100 μg/mL),resveratrol (100 μg/mL),psoralen (100 μg/mL),recipe water extract (100 μg/mL) and osthole (20,100 μg/mL) had a significant inhibition effects on the release of TNF-α;Ferulic acid (100 μg/mL),psoralen (100 μg/mL),resveratrol (20,100 μg/mL) and osthole (20,100 μg/mL) had significant inhibition effect on IL-6.Paeoniflorin (100 μg/mL),recipe water extract (100 μg/mL) and the other 4 components (20,100 μg/mL) had a remarkable inhibition effect on the release of NO.The results showed that the monomers hold generally stronger inhibition effect than recipe water extract.The mechanism of Bushen Huoxue recipe in treating osteoarthritis may be performed by inhibiting the release of inflammatory mediators of TNF-α,IL-6 and NO,which were helpful for exploring material basis of Bushen Huoxue recipe.
