美国肉参胶原蛋白肽对H2O2损伤PC12细胞的保护作用

Protective Effects of Collagen Peptide from Isostichopus badionotus on PC12 Cells Injured by H2O2

摘要: 采用风味蛋白酶对美国肉参体壁进行酶解,经超滤制备不同分子量的美国肉参胶原蛋白肽I1(6 kD<Mr<10 kD)和I2(Mr<6 kD)。利用H2O2诱导PC12细胞建立氧化损伤模型,比较I1和I2对氧化应激损伤的神经细胞的保护作用及机制。结果显示:不同浓度的I1、I2均能显著提高H2O2损伤的PC12细胞的增殖活性(P<0.05,P<0.01),降低细胞内ROS水平,减少LDH释放量和MDA含量(P<0.05,P<0.01),提高细胞的SOD、CAT和GSH-Px活力(P<0.05,P<0.01),抑制caspase-3酶活性的升高。表明美国肉参胶原蛋白肽具有良好的抗氧化活性,可以保护H2O2诱导的神经细胞损伤。其中,低分子量段的胶原蛋白肽组分保护效果更好。

Abstract: The body wall of Isostichopus badionotus was hydrolyzed with flavourzyme.Two fractions with different molecular weights I1 (6kD<Mr<10kD) and I2 (Mr<6kD) were obtained by ultrafiltration.The protective effects of I1 and I2 on H2O2-induced damage in PC12 cells were investigated.Results showed that all doses of I1 and I2 can promote the growth of PC12 injured by H2O2 obviously (P<0.05,P<0.01).The intracellular ROS,LDH release and the content of MDA decreased significantly (P<0.05, P<0.01).The activities of SOD,CAT and GSH-Px were stimulated remarkably (P<0.05,P<0.01).The caspase-3 activities decreased significantly (P<0.01).The collagen peptide with low molecular weight showed a better cytoprotective activity.These results demonstrated that collagen polypeptides from I.badionotus had remarkable antioxidant activity and can play a neuroprotective role in PC12 cells oxidative damaged by H2O2.
