高效液相色谱法同时测定青钱柳中9种指标成分的含量

Simultaneous Determination of Nine Chemical Markers of Cyclocaryae Folium by HPLC

摘要: 建立高效液相色谱法同时测定青钱柳中绿原酸、香草酸、逆没食子酸、异槲皮苷、阿福豆苷、槲皮苷、山奈酚、山奈素和乌苏酸含量的方法。采用Agilent ZORBAX Eclipse XDB-C18色谱柱(150 mm×4.6 mm,3.5 μm);流动相为0.2%磷酸乙腈-0.2%磷酸水,梯度洗脱;流速为0.7 mL/min,柱温35 ℃;检测波长:香草酸、逆没食子酸和乌苏酸为210 nm;绿原酸、异槲皮苷、阿福豆苷、槲皮素、山奈酚和山奈素为360 nm。青钱柳在该色谱条件下9种成分分离较好,平均加样回收率为95.86%~103.30%,RSD<2.5%。该方法简便、准确、重复性好,适用于青钱柳中绿原酸、香草酸、逆没食子酸、异槲皮苷、阿福豆苷、槲皮苷、山奈酚、山奈素和乌苏酸含量的同时测定。

Abstract: HPLC method was developed for the simultaneous determination of chlorogenic acid,vanililic acid,ellagic acid, sioquercitin, afzelin, quercetin, kaempferol, kaempferidel and ursolic acid in Cyclocaryae Folium.The ZORBAX Eclipse XDB-C18 column (150 mm×4.6 mm,3.5 μm) was used with 0.2% phosphoric acid in acetonitrile (A) and 02% phosphoric acid in water (B) as mobile phases in gradient elution mode.The flow rate was 0.7 mL/min,the column temperature was 35 ℃.The DAD detection wavelength was set at 210 nm for vanililic acid,ellagic acid and ursolic acid,360 nm for the others.As a result,nine chemical markers were separated well,the average recoveries were between 9586-103.30%,RSD<2.5%.The developed HPLC method was simple,accurate and reproductive.It is suitable for the determination of nine chemical markers of Cyclocaryae Folium.
