川黄柏果实提取物对肿瘤细胞的体外抑制作用及其机制

Proliferation Inhibition of Phellodendron chinense Schneid Fruit Extracts on Tumor Cells and Their Mechanisms

摘要: 研究川黄柏果实提取物的对体外癌细胞(HEL、K562、BPH1、PC3、MDA)生长的影响,探讨提取物对细胞生长的调控机制。采用MTT法检测川黄柏果实提取物对肿瘤细胞增殖的抑制作用,倒置荧光显微镜显微镜观察细胞形态及凋亡的变化,流式细胞术检测川黄柏果实提取物对细胞周期及细胞凋亡的影响。与对照组相比,川黄柏果实提取物HBE及匹西狄醇A(Piscidinol A)显著抑制BPH1细胞的增殖,但不能诱导细胞凋亡,显著缩短细胞G0/G1期(P<0.01),延长S期(P<0.01)。HBE、HBP及Piscidinol A能显著抑制HEL细胞的生长,轻微地诱导细胞凋亡,但对细胞周期影响不显著。

Abstract: To study the influence of extracts from the fruits of Phellodendron chinense Schneid in tumor cells(HEL,K562,BPH1,PC3,MDA) and to explore their effects on proliferation,cell cycle and apoptosis of tumor cells.The influence of extracts from the fruits of Phellodendron chinense Schneid in proliferation of HEL,K562,BPH1,PC3,MDA cells was measured by MTT assay.Cell cycle and apoptotic rate were analyzed by flow cytometry (FCM) in HEL and BPH1 cells.Compared with the control group,the HBE and Piscidinol A could inhibit the proliferation,shorten G0/G1 (P<005) obviously 〖JP3〗and extend S of BPH1cells,but they could not induce BPH1 cells’ apoptosis.The extracts (HBE,HBP and Piscidinol A) also could inhibit the growth and induce apoptosis of HEL cells,but they had no effect on cell cycle.
