超高效液相色谱法同时测定川射干中7个异黄酮成分的含量

Simultaneous Determination of 7 Isoflavone Components in Iris tectorum Using UPLC

摘要: 分析测定不同产地的川射干中射干苷、鸢尾甲苷A、鸢尾甲苷B、野鸢尾苷、鸢尾苷元、鸢尾甲黄素A、鸢尾甲黄素B 7个异黄酮成分,建立快速测定其异黄酮成分含量的方法。采用Waters Acquity UPLC系统;色谱柱为Waters BEH C18柱(2.1 mm×50 mm,1.7 μm);流动相为乙腈-0.5%冰醋酸,梯度洗脱;流速0.5 mL/min;柱温:35 ℃;检测波长266 nm。所得结果表明川射干中异黄酮成分分离良好,各成分进样量与峰面积在测定范围内呈现良好的线性关系(r≥0.9990),加样回收率分别为99.68%、100.57%、98.43%、98.03%、99.14%、9753%、101.86%,RSD 分别为0.84%、1.82%、1.67%、1.54%、0.89%、1.95%、1.77%。本实验所建立的川射干UPLC含量测定方法快速、简便、准确,为川射干的质量控制提供了科学依据。

Abstract: To establish a UPLC method for the simultaneous determination of tectoridin,iristectorin A,iristectorin B,iridin,tectorigenin,iristectorigenin A,iristectorigenin B in Iris tectorum Maxim..Waters BEH C18(2.1 mm×50 mm,1.7 μm) column was used,with acetonitrile-0.5% glacial acetic acid as the mobile phases in gradient elution mode.The column temperature was 35 ℃,with flow rate 0.5 mL/min.The UV detection wavelength was set at the maximum absorption wavelength,266 nm for all components.The seven kinds of isoflavone components in I.tectorum were separated well,and the linear relationship was obtained (r≥0.9990).The average recoveries were 99.68%,100.57%,98.43%,98.03%,99.14%,97.53% and 101.86%,with RSDs were 0.84%,1.82%,1.67%,1.54%,0.89%,1.95% and 1.77%,respectively.The established method was rapid,simple and accurate,it can provide the basis for the quality control of I.tectorum.
