大孔树脂纯化马甲子五环三萜类成分的工艺研究

Purification of Pentacyclic Triterpenoids from Paliurus ramosissimus by Macroporous Resin

摘要: 研究马甲子中五环三萜类化合物白桦脂酸、马甲子素的大孔吸附树脂纯化工艺。以白桦脂酸、马甲子素为指标,通过对10种大孔树脂(DA201、AB-8、D1400、D4020、NKA-9、HPD100、HPD700、H130和X-5)的静态、动态饱和吸附量和解析率实验筛选适宜的大孔树脂,并优化纯化工艺。工艺条件:马甲子素、白桦脂酸浓度分别为16.07、1023 μg/mL,上样体积为1000 mL(上样量为0.4 g/g,醇提物/干树脂),洗脱剂为85%乙醇,用量为900 mL,洗脱速度为3 mL/min。经树脂纯化后马甲子素的含量提高了8倍,白桦脂酸的含量提高了5倍,色素等杂质含量大大降低。大孔吸附树脂纯化后的马甲子提取物,其Hep-G2、AGS细胞的IC50分别为30.79±8.49、8.75±2.83 mg/L。本纯化工艺条件合理,适用于马甲子五环三萜类有效成分的纯化富集。

Abstract: To investigate the purification technology of pentacyclic triterpenoids from Paliurus ramosissimus by macroporous resin.The absorption and desorption capacity of 10 resin (DA201,AB-8,D1400,D4020,NKA-9,HPD100,HPD70,H130 and X-5) for betulinic acid and paliurusene were investigated to select the optimal macroporous resin.The adsorption characters and the influencing factors of the selected macroporous resin were also investigated to optimize the purification technology.The optimized conditions were as follows:the concentration of betulinic acid was 16.07 μg/mL,the concentration of paliurusene was 1023 μg/mL,the loading volume was 1000 mL (loading amount was 0.4 g/g),eluent was 85% ethanol,the amount of 900 mL,elution rate of 3 mL/min.After purification of the resin,the content of paliurusene was increased by 8 times,the content of betulinic acid was increased by 5 times,and the content of pigment and other impurities was greatly reduced.The IC50 of Hep-G2 and AGS cells were 30.79±8.49,8.75±2.83 mg/L,respectively,after purification using macroporous resin.The purification process was reasonable,suitable for the purification of active ingredients in Paliurus ramosissimus.
