山药中尿囊素对环磷酰胺诱导卵巢功能早衰的保护作用

Protective effect of allantoin from Dioscoreae Rhizoma on cyclophosphamide-induced premature ovarian failure in rats

摘要:

本文旨在探讨山药中尿囊素(allantoin,ALL)对卵巢功能早衰大鼠卵巢的保护作用及其作用机制。采用一次性腹腔注射环磷酰胺75 mg/kg对SD大鼠造成卵巢功能早衰模型,同时给予ALL进行干预,采用卵巢指数、病理切片中卵泡的发育与成熟以及血清中激素水平评估大鼠卵巢功能,运用比色法检测血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)与丙二醛(MDA)的水平,采用免疫组化与Western blotting检测卵巢组织中核因子E2相关因子2(Nrf2),血红素加氧酶1(HO-1)和NAD(P)H醌脱氢酶1(NQO1)蛋白表达。结果显示:与正常组(NC)相比,卵巢早衰模型组(POF)大鼠卵巢指数与体内雌二醇(E2)水平降低(P<0.05或P<0.01),黄体生成素(LH)与促卵泡生成素(FSH)水平升高(P<0.05或P<0.01),卵巢组织中初级,次级以及成熟卵泡减少,闭锁卵泡增加,卵巢组织匀浆中SOD、GSH-Px、Nrf2与HO-1水平显著降低(P<0.05或P<0.01),而MDA水平显著升高(P<0.05);治疗14 d后,与POF组相比,ALL可以升高血清中E2,降低LH与FSH水平(P<0.05或P<0.01),增加卵巢组织中的初级、次级以及成熟卵泡,升高SOD、GSH-Px、Nrf2、NQO1与HO-1水平(P<0.05或P<0.01),降低MDA水平(P<0.05),从而改善氧化应激造成的卵巢损伤。山药中尿囊素可以通过Nrf2/HO-1途径改善卵巢功能早衰大鼠的氧化应激,对大鼠的卵巢具有保护作用。

Abstract:

The objective of this study was to investigate the protective effect and the mechanism of allantoin (ALL) from Dioscoreae Rhizoma on ovaries of rats with premature ovarian failure.A one-time intraperitoneal injection of 75 mg/kg cyclophosphamide was used to create a model of premature ovarian failure in SD rats,and ALL was administered.Ovarian index,follicle development in pathological sections and serum hormone levels were used to assess the ovarian function of the rats.Superoxide dismutase (SOD),glutathione peroxidase(GSH-Px) and malondialdehyde (MDA) levels of ovary were detected by colorimetry.Immunohistochemistry and Western blotting were used to detect nuclear factor erythroid-2 related factor 2 (Nrf2),heme oxygenase-1(HO-1) and NAD(P)H quinone dehydrogenase 1(NQO1) protein expressions.The results show,compared with the nomal contol (NC) group,the ovarian index and the level of estradiol (E2) in POF group were decreased (P<0.05 or P<0.01),the levels of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) were increased (P<0.05 or P<0.01),the primary,secondary and mature follicles in ovarian tissue were decreased,and the atretic follicles were increased.SOD and GSH-Px,Nrf2 and HO-1 levels in tissue homogenate were significantly decreased (P<0.05 or<0.01),while the MDA level was significantly increased (P<0.05).After 14 days of treatment,compared with POF group,ALL increased the level of E2,decreased the levels of LH and FSH in the POF rats(P<0.05 or P<0.01),and increased primary,secondary and mature follicles in ovarian tissue of POF rats.The levels of SOD,GSH-Px,Nrf2,NQO1 and HO-1 were increased (P<0.05 or P<0.01),and the levels of MDA were decreased (P<0.05).Allantoin from Dioscoreae Rhizoma modifies oxidative stress in rats with premature ovarian failure through Nrf2/HO-1 pathway,and has protective effect on the ovaries of rats.
