银线草地上部分化学成分及其化疗增敏作用研究

Chemical constituents from the aerial parts of Chloranthus japonicus and their chemosensitizing activities

摘要: 研究银线草地上部分化学成分,并考察其对人骨肉瘤U2OS细胞化疗增敏性的影响。采用硅胶、Toyopearl HW-40C、ODS柱色谱及半制备液相等方法对银线草地上部分的石油醚部位进行分离纯化,根据理化性质、波谱数据和ECD计算,并结合文献比对鉴定化合物结构。从中共分离鉴定了13个化合物,分别为chlojaparomatic(1)、银线草醇B(2)、银线草醇F(3)、chlorajaponol(4)、银线草醇C(5)、银线草醇E(6)、sarglabolide I(7)、cycloshizukaol A(8)、chloranholosin B(9)、(–)-loliolide(10)、3-isopropyl-5-acetoxycyclohexene-2-one-1(11)、2'-羟基-4,3',4',6'-四甲氧基查尔酮(12)和5,7,4'-trimethoxyflavanone(13)。其中,化合物1为新化合物;9~13为首次从该植物中分离得到。生物活性评价结果显示,二氯甲烷部位对人乳腺癌MCF-7细胞有较好抑制作用。石油醚部位、二氯甲烷部位在50、100 μg/mL浓度下对阿霉素抗U2OS细胞具有协同增效作用;化合物1可明显提高阿霉素对U2OS细胞的敏感性,其50 μmol/L浓度下的联合指数为0.77 ± 0.40。

Abstract:

This study aims to investigate the chemical components of the aerial parts of Chloranthus japonicus and their chemosensitizing activities on human osteosarcoma U2OS cells. Thirteen compounds were isolated and purified from petroleum ether extract of the aerial parts of C. japonicus by column chromatography over silica gel, Toyopearl HW-40C, ODS and semi-preparative high performance liquid chromatography. Their chemical structures were elucidated based on physiochemical properties, spectral datas, ECD calculation and literature comparisons, as chlojaparomatic (1), shizukaol B (2), shizukaol F (3), chlorajaponol (4), shizukaol C (5), shizukaol E (6), sarglabolide I (7), cycloshizukaol A (8), chloranholosin B (9), (–)-loliolide (10), 3-isopropyl-5-acetoxycyclohexene-2-one-1 (11), 2'-hydroxy-4,3',4',6'-tetramethyl chalcone (12) and 5,7,4'-trimethoxyflavanone (13). Among them, compound 1 was one new compound; and 9-13 were isolated from this species for the first time. Bioassay results showed that the dichloromethane extract had an inhibitory effect on human breast cancer MCF-7 cells. The petroleum ether and dichloromethane extracts at 50 and 100 μg/mL had synergistic cytotoxic effects in combination with doxorubicin on U2OS cells. Compound 1 increased the sensitivity of U2OS cells to doxorubicin, and the combination index at 50 μmol/L was 0.77 ± 0.40.
