文冠果种粕化学成分及其抗炎活性研究

Chemical constituents from Xanthoceras sorbifolia seed meal and their anti-inflammatory activity

摘要:

研究文冠果种粕的化学成分及其抗炎活性。采用重结晶、Sephadex LH-20凝胶柱色谱、硅胶柱色谱及半制备高效液相色谱等方法对文冠果种粕乙醇提取物正丁醇萃取部位进行分离纯化,并根据理化性质、核磁及质谱数据鉴定所得化合物的结构。通过Griess法检测提取物及部分化合物对脂多糖诱导的RAW 264.7细胞产生NO的抑制作用。从文冠果种粕乙醇提取物中分离到20个化合物,分别为儿茶酚(1)、阿魏酸(2)、咖啡酸(3)、咖啡酸乙酯(4)、对甲氧基肉桂酸乙酯(5)、邻苯二甲酸二(2-乙基己基)酯(6)、邻苯二甲酸二丁酯(7)、邻苯二甲酸丁酯异丁酯(8)、对苯二甲酸二丁酯(9)、秦皮甲素(10)、爵床脂素A(11)、aneglycoside D(12)、尿素(13)、5-octenyl sulfate(14)、二十八烷醇(15)、赤藓醇(16)、乳酸醚(17)、(Z)-2-丁氧基-8-十七碳烯(18)、6, 9-十七二烯(19)、蔗糖(20),均是首次从文冠果中分离得到。体外抗炎活性结果表明,正丁醇萃取部位(300 μg/mL)以及化合物8(200 μmol/L)、10(200 μmol/L)、14(200 μmol/L)均能显著抑制脂多糖诱导的RAW 264.7细胞释放NO,抑制率分别为22.07%、17.65%、30.98%、21.15%,具有潜在抗炎活性。

Abstract:

This study aims to investigate the chemical constituents and anti-inflammatory activity of Xanthoceras sorbifolia seed meal. The n-butanol extract of the ethanol extract of X. sorbifolia seed meal was isolated and purified by recrystallization, Sephadex LH-20 gel column chromatography, silica gel column chromatography and semi-preparative high performance liquid chromatography. The inhibitory effects of the extract and selected compounds on NO production in LPS-induced RAW 264.7 cells were detected using the Griess method. The structures of the obtained compounds were identified by physicochemical properties, nuclear magnetic resonance and mass spectrometry data. Twenty compounds were isolated from the ethanol extract of X. sorbifolia seed meal and identified as catechol (1), ferulic acid (2), caffeic acid (3), ethyl caffeate (4), 4-methoxycinnamic acid ethyl ester (5), bis(2-ethylhexyl) phthalate (6), dibutyl phthalate (7), butylisobutyl phthalate (8), dibutyl terephthalate (9), esculin (10), justicidin A (11), aneglycoside D (12), carbamide (13), 5-octenyl sulfate (14), octacosanol (15), erythritol (16), dilactic acid (17), (Z)-2-butyroxy-8-heptadecene (18), 6, 9-heptadecadiene (19) and sucrose (20). All compounds were isolated from X. sorbifolia for the first time. The results of anti-inflammatory activity in vitro showed that the n-butanol fraction (300 μg/mL) and compounds 8 (200 μmol/L), 10 (200 μmol/L) and 14 (200 μmol/L) could significantly inhibit the release of NO from RAW 264.7 cells induced by lipopolysaccharide, and the inhibition rates were 22.07 %, 17.65 %, 30.98 % and 21.15 %, respectively, with potential anti-inflammatory activity.
