藏药铁棒锤的二萜生物碱成分及其抗炎活性研究

Diterpenoid alkaloids from Aconitum pendulum Busch and their anti-inflammatory activities

摘要:

研究藏药铁棒锤(Aconitum pendulum Busch.)中二萜生物碱化学成分及其抗炎活性。综合运用硅胶吸附柱色谱和薄层色谱等多种色谱技术对铁棒锤块根中的生物碱类成分进行分离纯化,根据波谱解析及理化性质鉴定所得化合物的结构,采用MTT法和Griess法测定化合物的细胞毒性和抗炎活性。从铁棒锤块根的乙醇提取物中分离鉴定出13个二萜生物碱类化合物,分别为新乌头碱(1)、次乌头碱(2)、宋果灵(3)、3-脱氧乌头碱(4)、乌头碱(5)、3-乙酰乌头碱(6)、2-羟基脱氧乌头碱(7)、8-O-甲基乌头碱(8)、3-脱氧乌头原碱-8-亚油酸酯(9)、14-苯甲酰基乌头原碱-8-油酸酯(10)、14-苯甲酰基乌头原碱-8-棕榈酸酯(11)、14-苯甲酰基乌头原碱-8-亚油酸酯(12)、azitine(13),其中化合物910~13为首次铁棒锤生品中分离得到。体外抗炎活性结果表明,化合物1~69~12均能够抑制RAW 264.7细胞中NO的生成,具有一定的抗炎活性,其中化合物459~12通过降低RAW 264.7细胞中炎症因子白细胞介素-1β(interleukin-1β,IL-1β)、IL-6、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的水平发挥抗炎作用,化合物12抗炎作用较强,具有进一步深入研究的价值。

Abstract:

This study aims to investigate the chemical constituents of diterpenoid alkaloids from the roots of Aconitum pendulum Busch. and their anti-inflammatory activities. The compounds were isolated and purified by using silica gel column chromatography and thin layer chromatography, and the structures of the compounds were identified by spectral analysis and physicochemical properties. Meanwhile, the MTT and Griess methods were used to evaluate the anti-inflammatory activities of compounds. A total of 13 diterpenoid alkaloids compounds were identified as mesaconitine (1), hypaconitine (2), songorine (3), 3-deoxyaconitine (4), aconitine (5), 3-acetylaconitine (6), 2-hydroxydeoxyaconitine (7), 8-O-methylaconitine (8), 3-deoxyaconine-8-linoleate (9), 14-benzoylaconine-8-oleate (10), 14-benzoylaconine-8-palmitate (11), 14-benzoylaconine-8-linoleate (12), azitine (13). Compounds 9, 10-13 were isolated from the roots of A. pendulum for the first time. Compounds 1-6, 9-12 all showed inhibition effects on NO production in RAW 264.7 cells and have certain anti-inflammatory activity. ELISA assays revealed that compounds 4, 5, 9-12 could reduce the expression of inflammatory cytokines interleukin-1β (IL-1β), IL-6 and tumor necrosis factor-α (TNF-α). Compound 12 has a relatively strong anti-inflammatory effect, suggesting its potential value for further in-depth research.
