白囊耙齿菌多糖结构及抗肾小球系膜细胞的增殖活性研究

Structure of polysaccharide and its resistance to GMC proliferation activity from Irpex lacteus

摘要: 为表征白囊耙齿菌多糖结构并筛选出具有抗肾小球系膜细胞(GMC)增殖作用的活性多糖,研究活性多糖对GMC异常增殖及细胞外基质分泌的影响。实验采用碱提醇沉法提取粗多糖(PC),经离子交换色谱柱纯化、SePhadex G50凝胶柱分离得到耙齿菌多糖PCP1、PCP2。通过苯酚硫酸法、BCA法等测定其含量,甲基化,IR分析进行结构表征。采用MTT法、ELISA法考察对GMC增殖的影响。结果表明,PCP1和PC2的总糖含量分别为98.05%、96.92%,酸性糖及蛋白质含量几乎为零; PCP1主要由Man、Glc和Gal组成,具有多分枝结构,包含吡喃糖类型的结构,具备αβ两种构型;而PCP2仅由Glc组成,没有分支,为α构型吡喃聚糖。具有抗GMC增殖活性的均一多糖为PCP1,可保护LPS诱导的GMC增殖,减少LN、FN蛋白的释放。

Abstract: To characterize the polysaccharide structure of Irpex lacteus and screen active polysaccharides with antiproliferation effect on glomerular mesangial cells (GMC),the effects of active polysaccharides on abnormal GMC proliferation and extracellular matrix secretion were studied.Crude polysaccharide (PC) was extracted by alkali extraction and alcohol precipitation method,and PCP1 and PCP2 were obtained by purification by ion exchange chromatography column and separation by sephadex g50 gel column.The physical and chemical properties,methylation and IR analysis of the polysaccharides were determined by phenol sulfuric acid method and BCA method.MTT assay was used to investigate the effect on the proliferation of glomerular mesangial cells (GMC).As a result,the total sugar content of PCP1 and Pc2 was 98.05% and 96.92%,respectively.PCP1 is mainly composed of Man,Glc and Gal,and the Gal content is the highest.While PCP2 only consists of Glc,the homopolysaccharide with antiGMC proliferative activity is PCP1,which can protect the GMC valueadded induced by LPS and reduce the release of LN and FN proteins.
