青藤碱对MsPGN大鼠肾脏病理及ICAM-1表达的影响

Affection of Sinomenine on the Kidney Pathology and Expression of ICAM-1 in the Rats with Mesangial Proliferative Glomerulonephritis

摘要:

为探讨青藤碱(Sinomenine,SIN)对实验性系膜增生性肾小球肾炎(MsPGN)的病理形态学改善及肾组织中细胞间粘附分子-1(ICAM-1)表达的影响,通过实验建立改良的慢性血清病性MsPGN动物模型,光镜观察肾小球以及肾小管-间质的病理改变情况,采用免疫组织化学法检测ICAM-1的表达并分别进行半定量分析。结果显示:光镜下,模型组与正常组相比,系膜基质指数显著升高(P<0.01),肾小球毛细血管直径明显缩小(P<0.01);青藤碱组与模型组相比,上述病理改变明显减轻,系膜基质指数显著下降(P<0.01),肾小球毛细血管直径明显改善(P<0.01)。肾组织中ICAM-1免疫组化结果显示青藤碱可明显 下调ICAM-1的表达,肾组织ICAM-1的相对含量、积分光密度,均显著下降(P<0.01),青藤碱组与雷公藤多苷组相比,二者无统计学意义(P >0.05)。实验表明青藤碱能够有效减轻肾脏病理损害,抑制MsPGN大鼠肾组织ICAM-1的表达,延缓疾病进展。

Abstract:

To explore affection of Sinomenine (SIN) on the kidney pathology and Intercellular cell adhesion molecule-1 (ICAM-1) expression in rats with mesangial proliferative glomerulonephritis (MsPGN).The test uses the improvement chronic serum sickness nature MsPGN model,with the light mirror observing mesangial region and kidney tubules-mesenchyma,using immunohistochemical method observe the expression of ICAM-1,at the same time,carries on the semi-definite quantity analysis with the pathology image analysis system.The results showed that compared with the normal group,mesangial matrix index (%) remarkably heighten (P<0.01) and capillary lumen diameter (μm) obviously reduce.Compard with the model group,in SIN group,mesangial matrix index(%)remarkably reduce (P<0.01) and capillary lumen diameter (μm) obviously improve (P<0.01).In the nephridial tissue immunohistochemistry result showed that SIN can obviously reduce Relative amount (%) and Density expression (P<0.01).Comparing between two treatments groups,although the result of SIN group is lower than TWP group but the two comparison have no the remarkable difference (P>0.05).This result suggested that SIN can effectively reduce the kidney pathological lesion,refrain the expression of ICAM-1.
