肾络通对系膜增生性肾小球肾炎大鼠血脂代谢及肿瘤坏死因子表达的影响

Effect of Shenluotong on Lipid-metabolism and TNF-α Expression in Rats with Mesangial Proliferative Glomerulonephritis

摘要: 探讨中成药肾络通胶囊对实验性系膜增生性肾小球肾炎(MsPGN)的血脂代谢及肿瘤坏死因子-α(TNF-α)表达的影响。采用改良的慢性血清病性MsPGN模型,检测血清TC、TG、HDL、LDL及采用免疫组织化学法和逆转录多聚酶链反应(RT-PCR)技术检测TNF-α及TNF-α mRNA的表达并进行半定量分析。结果显示两治疗组血清TC、TG、LDL明显降低(P<0.01,P <0.05),HDL则显著升高(P<0.01)。肾络通组TC略高于雷公藤多甙组,TG、HDL略低于雷公藤多甙组,LDL与雷公藤多甙组相等,但两治疗组相比无统计学意义(P>0.05)。肾组织中TNF-α免疫组化及RT-PCR结果显示肾络通可有效下调TNF-α及TNF-α mRNA的表达,与模型组相比有显著差异(P<0.01,P<0.05),肾络通组与雷公藤多甙组相比,二者无统计学意义(P>0.05)。从以上结果说明肾络通能够有效改善高血脂,抑制肾小球分泌TNF-α及下调TNF-α mRNA的表达,延缓疾病进展。

Abstract: To explore the effect of Shenluotong (SLT) on lipid metabolism and TNF-α expression in rats with mesangial proliferative glomerulonephritis (MsPGN),the improved chronic serum sickness nature MsPGN model was made and TC,TG,HDL,LDL were assayed with automatic biochemistry analyzer.At the same time,the expression of TNF-α and TNF-α mRNA was observed and analyzed with the semi-definite quantity analysis with immunohistochemistry staining and reverse transcription polymerase chain reaction (RT-PCR).The results showed the serum TC,TG,LDL in two treatment groups were decreased significantly (P<0.01, P<0.05). HDL was increased significantly (P<0.01).TC in SLT was significantly higher (P<0.01) than that of Tripterygium wilfordii (TW) group.TG,HDL were slightly lower than that of TW group.LDL was equivalent in the two treatment groups,but there was no statistical significance between SLT and TW group (P<0.01).SLT can down-regulate the expression of TNF-α and TNF-α mRNA in kidney and there were significant differences between SLT group and model group (P<0.01,  P<0.05), there was no statistical significance between two treatment groups (P>0.05).These results suggested that SLT can effectively improve high blood lipid,and played a role in inhibiting the secretion of TNF-α and the expression of TNF-α mRNA in glomerulus.Hence,SLT can delay the progression of the disease.
