北虫草体外抗氧化和PC12氧化损伤修复作用的有效部位的筛选

Antioxidant Activity and Oxidative Injury Protection for PC12 of Three Extracts from Cordyceps militaris Fruiting Bodies in vitro

摘要: 采用系统溶剂法对北虫草子实体进行依次提取,制备得氯仿相、乙酸乙酯相和乙醇相三部位,利用化学发光法和H2O2诱导PC12氧化损伤修复模型,对三部位进行体外抗氧化活性和PC12氧化损伤修复作用测定。结果表明,乙酸乙酯相具有较强的抗氧化活性,是清除H2O2自由基和超氧自由基活性最强的部位,IC50值分别为88.5 µg/mL和190 µg/mL;乙酸乙酯相对由H2O2诱导的PC12细胞氧化损伤的修复作用较强,且呈现明显的浓度依赖性。无论从抗氧化活性,还是从对H2O2氧化损伤的修复作用,乙酸乙酯相均表现出了较强的作用,乙酸乙酯相是抗氧化活性和保护PC12细胞氧化损伤的主要有效部位。

Abstract: Cordyceps militaris fruiting bodies were extracted by using the method of systematic solvent extraction, and three different extracts were prepared named chloroform extract, acetic ether extract and ethanol extract, respectively. In vitro, the antioxidant activities of different extracts were investigated by using chemiluminescence measurements, and the activities of oxidative injury protection were assessed by using PC12 oxidative injury model induced with H2O2. The superoxide free radical (O2-) system and H2O2 system assays showed that the acetic ether extract had notable antioxidant activity, their scavenging rate for both H2O2 and O2- were highest among the three extracts, whose IC50 were 88.5 µg/mL and 190 µg/mL, respectively. Moreover, the oxidative injury protection for PC12 assay showed that the acetic ether extract had significant rehabilitation effect in comparison with chloroform extract and ethanol extract, and it rehabilitated PC12 cell treated with H2O2 in a dose-dependent manner. Therefore, it was concluded that the acetic ether extract was the principal extract of both antioxidant and the activity of oxidative injury rehabilitation among three extracts from C. militaris fruiting bodies.
