HPLC法同时测定新疆圆柏叶中芦丁、异槲皮苷及槲皮苷的含量

Simultaneous Determination of the Leaves of Rutin,Isoquercitrin and Quercetin Content in the Leaves of Juniperus sabina L.by HPLC

摘要: 建立高效液相色谱法同时测定新疆圆柏叶中芦丁、异槲皮苷及槲皮苷含量的方法。采用Phenomenex GeminiNX C18:色谱柱(250×4.6 mm,5 μm),以乙腈(A)0.2%磷酸水溶液(B)为流动相梯度洗脱,柱温30 ℃,流速1.0 mL/min,检测波长360 nm。新疆圆柏叶中芦丁、异槲皮苷及槲皮苷在25 min内分离效果理想,线性范围分别为10.4~62.4 μg/mL(r=0.9999),3.0~18.0 μg/mL(r=0.9998),11.0~66.0 μg/mL (r=0.9999);平均加样回收率分别为98.64%、100.80%和98.81%。该方法简单易行,能够控制新疆圆柏叶药材的质量标准。将该方法应用于研究不同采收期对含量的影响,结果显示在新疆圆柏生长最旺盛的7月底,这3个黄酮类成分的含量最高。

Abstract: To establish an efficient high performance liquid chromatography(HPLC)method for the simultaneous determination of rutin,isoquercitrin and quercetin in the leaves of Juniperus sabina L.Samples were analyzed on a Phenomenex GeminiNX C18∶〖KG-*2〗 column(250×4.6 mm,5 μm)eluted with acetonitrile(A)and 0.2% phosphoric acid solution(B)as the mobile phase in a gradient model;the column temperature was 30 ℃,the flow rate was 2.0mL/min,the detective wavelength was 360 nm.Rutin,isoquercitrin and quercetin in the leaves of Juniperus sabina L.were ideally separated in 25 min,and their linear response ranges for them were 10.4~62.4 μg/mL(r=0.9999),3.0~18.0 μg/mL(r=0.9998)and 11.0~66.0 μg/mL(r=0.9999),respectively.The average recovery rate was 98.64%,100.80% and 98.81% respectively.This method is simple,practicable and can be used as the quality control method of Juniperus sabina L.This method has been successfully applied to the analysis in different gathering periods of Juniperus Sabina L.The result showed that content of three components in Juniperus sabina were highest in the end of July.
