摘要:

该研究旨在通过液质联用技术鉴定人参发酵液(Panax ginseng fermentation broth,GFB)中人参皂苷成分,结合网络药理学、分子对接和实验验证探究GFB美白活性及作用机制。结果表明,GFB共含有15种主要人参皂苷成分,网络药理学和分子对接预测GFB主要通过作用于高级糖基化终产物-受体/小眼畸形相关转录因子-酪氨酸酶(advanced glycation end products-receptor for advanced glycation end products/ microphthalmia-associated transcription factor-tyrosinase,AGE-RAGE/MITF-TYR)通路,调节酪氨酸酶的表达和细胞对氧化应激的反应,从而减少黑色素合成。细胞实验结果表明,GFB除了能显著降低细胞活性氧(reactive oxygen species,ROS)积累(P < 0.001),还能显著抑制B16小鼠黑色素瘤细胞中黑色素含量和酪氨酸酶活性(P < 0.05)。GFB通过抑制AGE-RAGE/MITF-TYR信号通路,降低皮肤内ROS积累和抑制酪氨酸酶活性,最终发挥美白活性,具有多成分-多靶点-多途径的特点。

Abstract:

The aim of this study is to identify the ginsenoside components in Panax ginseng fermentation broth (GFB) by liquid chromatography-mass spectrometry technology,and to investigate the whitening activity and mechanism of GFB by combining network pharmacology,molecular docking and experimental validation.The results showed that GFB contained 15 major ginsenoside components.Network pharmacology and molecular docking predicated that GFB reduced melanin synthesis mainly by acting on the advanced glycation end products-receptor for advanced glycation end products/microphthalmia-associated transcription factor-tyrosinase (AGE-RAGE/MITF-TYR) pathway,modulating tyrosinase expression and cellular response to oxidative stress.The results of cellular experiments showed that GFB significantly inhibited melanin content and tyrosinase activity in B16 mouse melanoma cells (B16) (P < 0.05),in addition to significantly reducing reactive oxygen species (ROS) accumulation in cells (P < 0.001).By inhibiting AGE-RAGE/MITF-TYR signaling pathway,GFB reduced ROS accumulation and inhibited tyrosinase activity in the skin,and ultimately exerted whitening activity with multi-component-multi-target-multi-pathway characteristics.
