紫萼玉簪花总皂苷的纯化及抗肿瘤活性研究

摘要: 本文旨在研究紫萼玉簪花总皂苷的纯化工艺及抗肿瘤活性。以比吸附量和比洗脱量为指标,筛选大孔树脂型号,进一步优化工艺条件。通过体外实验(MTT法)对紫玉簪花总皂苷进行抗肿瘤活性研究。实验结果显示AB-8型大孔树脂适合富集纯化紫萼玉簪花总皂苷,最佳纯化条件为药材量与树脂用量比小于1∶15,分别用4 BV水和20%乙醇除杂,6 BV 70%乙醇在2 BV·h-1流速下洗脱,在此条件下得到的紫萼玉簪花总皂苷(纯度57.49%)对SGC-7901、MCF-7、HepG-2肿瘤细胞有较强的抑制作用,IC50分别为15.47 μg·L-1、28.08 μg·L-1、17.37 μg·L-1,表明具有良好的抗肿瘤活性。

Abstract: The aim of this paper was to study the purification process of total saponins of H.ventricosa(TSHV) and their anti-tumor activities.Filtrate macroporous resin models and further optimize the process conditions with the adsorption ratio and elution ratio as an indicator.The purified TSHV were screened for antitumor activity in vitro by MTT assay.The results showed that AB-8 macroporous resin was suitable for the purification of TSHV.The optimal purification conditions were as follows: the ratio of H.ventricosa dosage to the amount of macroporous resin was less than 1∶15;4 BV water and 20% ethanol were used to remove impurities,and 6 BV 70% ethanol was eluted at 2 BV·h-1flow rate.Under these conditions,the purity of TSHV obtained was 57.49%.The prepared TSHV could effectively inhibit the proliferations of SGC-7901,MCF-7 and HepG-2 tumor cells in vitro,the IC50 values of which were 15.47 μg·L-1,28.08 μg·L-1 and 17.37 μg·L-1 respectively,which showed brilliant anti-tumor effects.
