黄海棠内生真菌Phomopsis prunorum次级代谢产物及其α-葡萄糖苷酶抑制活性研究

摘要:

本研究采用多种色谱技术从黄海棠内生真菌Phomopsis prunorum(F4-3)大米发酵产物中获得6个次级代谢产物,经核磁、质谱等波谱学方法对这些化合物进行了结构鉴定,分别为α-吡喃酮类化合物phomopyrones B、C(12)、二苯并α-吡喃酮类化合物alternariol和alternariol 9-methyl ether(34)、异香豆素类化合物(+)-orthosporin(5)和生物碱类化合物3-(2-(4-hydroxyphenyl)-2-oxoethyl)-5,6-dihydropyridin-2(1H)-one(6)。化合物1和2为新化合物,化合物4~6为首次从该属真菌中分离获得。药理活性评价显示,化合物3显示中等的α-葡萄糖苷酶抑制活性,其IC50值为43.4 μM;而化合物1、24~6均未见明显的α-葡萄糖苷酶抑制及抗菌活性。

Abstract:

Six secondary metabolites were obtained from the rice fermentation extract of endophytic fungus Phomopsis prunorum isolated from Hypericum ascyron L. by various chromatographic techniques.Their structures were elucidated by the analysis of NMR and MS data,including α-pyrone compounds phomopyrones B and C (1 and 2),dibenzo-α-pyrone compounds alternariol (3) and alternariol 9-methyl ether (4),isocoumarin compound (+)-orthosporin (5),and alkaloid compound 3-(2-(4-hydroxyphenyl)-2-oxoethyl)-5,6-dihydropyridin-2(1H)-one (6).Compounds 1 and 2 were new compounds,and compounds 4-6 were isolated from the genus Phomopsis for the first time.The biological activity results showed that compound 3 exhibited moderate α-glucosidase inhibitory activity with an IC50 value of 43.4 μM.However,compounds 1,2 and 4-6 showed no α-glucosidase inhibitory and antibacterial activities.
