Structural characterization and anti-inflammatory activity of neutral homogeneous polysaccharides from Prinsepia utilis oil meal

摘要:

对青刺果(Prinsepia utilis Royle)油粕中分离纯化得到的中性均一多糖进行结构表征及并研究其抗炎活性。通过水提醇沉法得到青刺果油粕粗多糖,经DEAE-52纤维素阴离子交换色谱柱和Sephadex G100凝胶过滤色谱柱纯化,得到青刺果油粕均一多糖(P. utilis oil meal polysaccharides-A1,PURP-A1);通过高效凝胶渗透色谱、离子色谱、傅里叶变换红外光谱、气相色谱-质谱联用及核磁共振等谱学技术对PURP-A1的相对分子质量、单糖组成、特征官能团、糖苷键类型及其连接方式进行解析。利用P物质诱导的人永生化角质形成细胞(HaCaT)炎症模型,评价其抗炎活性。结果显示,PURP-A1的相对分子质量为18 373 Da,主链由葡萄糖聚合而成,1条支链由阿拉伯糖聚合而成,二者通过α-糖苷键相连。500及1 000 μg/mL PURP-A1可显著降低P物质诱导的HaCaT细胞炎症模型中肿瘤坏死因子-α(tumor necrosis factor-αTNF-α)、白细胞介素-6(interleukin-6,IL-6)、IL-8、干扰素-γ(interferon-γIFN-γ)的mRNA表达。提示该多糖在神经源性炎症导致的皮肤疾病方面具有优良的治疗潜力,为该植物资源在皮肤健康领域的深度开发与科学应用提供了重要科学依据。

Abstract:

This study aims to investigate the structural characteristics and anti-inflammatory activity of neutral homogeneous polysaccharides isolated and purified from Prinsepia utilis Royle oil meal. The crude polysaccharides from P. utilis oil meal were extracted by water extraction and alcohol precipitation, followed by purification using DEAE-52 cellulose anion exchange chromatography and Sephadex G100 gel filtration chromatography to obtain P. utilis oil meal polysaccharides-A1 (PURP-A1), which was a homogeneous polysaccharide. The structural features of PURP-A1, including its relative molecular weight, monosaccharide composition, characteristic functional groups, glycosidic bond types, and linkage patterns, were analyzed using spectroscopic techniques such as high-performance gel permeation chromatography, ion chromatography, fourier transform infrared spectroscopy, gas chromatography-mass spectrometry, and nuclear magnetic resonance. The anti-inflammatory activities were evaluated using a substance P-induced immortalized human keratinocyte cells (HaCaT) inflammation model. The results showed that PURP-A1 exhibited a relative molecular weight of 18 373 Da, with the main chain polymerized by glucose residues and a branch chain polymerized by arabinose, which were connected to α-glycosidic bonds. At concentrations of 500 and 1 000 μg/mL, PURP-A1 significantly downregulated the mRNA expression of inflammatory cytokines tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), IL-8 and interferon-γ (IFN-γ) in substance P-induced HaCaT cells, demonstrating its therapeutic potential for neurogenic inflammatory skin diseases, providing an important scientific basis for the in-depth development and scientific application of this plant resource in the field of skin health.
