太子参细胞毒活性部位HPLC谱效关系初探

Correlation between Cytotoxic Activity and HPLC Fingerprint Chromatogram of the Effective Fractions of Radix Pseudostellariae

摘要: 采用MTT法观察太子参提取物对人胃癌MGC80-3、人结肠癌RKO和人肝癌HepG2细胞的抑制作用;运用TLC法检识太子参的环肽类化合物;分析太子参有效部位HPLC指纹图谱与细胞毒活性的谱效关系。结果表明,太子参乙酸乙酯部位对MGC80-3、RKO细胞具有抑制作用,进一步分离得到的组分G对三种肿瘤细胞均具有明显抑制作用,并比较两者的TLC和HPLC图谱,可知太子参有效部位的细胞毒活性是各特征峰成分共同作用的结果,并可能与环肽类化合物有关。

Abstract: MTT assay was applied to screen the cytotoxic activities of the fractions of Radix Pseudostellariae (RP) on cell lines of MGC80-3, HepG2 and RKO. Cyclopeptides of RP were identified by TLC. And the correlation between cytotoxic activity and the HPLC fingerprints of the effective fractions was analyzed. The results showed that the ethyl acetate fraction had inhibitory effect on MGC80-3 and HepG2 cell lines. Furthermore, the fraction G which isolated from the ethyl acetate fraction exhibited significant cytotoxicity against all these three tumor cell lines. Comparison the HPLC and TLC fingerprints of the ethyl acetate fraction with those of fraction G suggested that the coefficient of all characteristic constituents resulted in their cytotoxic effect. In addition, the effect may be correlated with the existence of cyclopeptides.
