Study on separation method of alkynysides in Codonopsis Radix and its hypoglycemic activity in vitro

Abstract:

This study confirmed that the chloroform methanol part of Codonopsis Radix (5 mg/mL) had good α-glucosidase inhibitory activity in vitro,the main component is alkynosides,the inhibition rate in vitro is more than 50%,and has good hypoglycemic activity in vitro.Taking the glycosides from Codonopsis Radix as the research object,combined with the UV spectral data of the compounds,construct an efficient and systematic preparation method for reversed-phase two-dimensional liquid chromatography and systematically separate it.The chloroform methanol part of Codonopsis Radix was obtained by extraction,sugar removal by D-101 macroporous resin column and chromatographic separation by silica gel adsorption column.using reverse column C18 (4.6 mm × 250 mm,5 μm) as a one-dimensional and two-dimensional separation column,acetonitrile and water as mobile phase,finally five alkynyside monomer compounds with purity greater than 95% were separated by semi-prepared high liquid chromatography,and the compound structure was determined according to the physicochemical properties and spectral data and identidentified as pratialin B,pilosulyne E,corfifolioidyne,lobetyolinin,lobetyolin.
