Optimization of ultrasound-assisted enzymatic extraction of lacquer wax and its physicochemical property

Abstract:

In this experiment,ultrasound-assisted enzyme method was used to extract lacquer wax from lacquer seeds,and the composition of its fatty acid composition was also identified in order to realize comprehensive utilization of lacquer seed resources.Firstly,this experiment has observed the effects of enzyme dosage,ultrasonic power,ultrasonic time,ultrasonic temperature,enzymatic hydrolysis time and pH on the extraction rate of lacquer wax,to further response the extraction technique parameters faced with experiment design optimization lacquer wax.Gas chromatography-mass spectrometer was used to analyze the compositions and relative contents of lacquer wax,and its physicochemical property were determination.The results showed that the optimum extraction parameters were as follows:enzyme dosage was 2.5%,enzymatic hydrolysis time was 50 min,ultrasonic power was 225 W,ultrasonic time was 25 min,ultrasonic temperature was 45 ℃,pH 4.5,enzymatic hydrolysis time was 50 min.Under this condition,the extraction rate of lacquer wax was 33.93%.The composition and physicochemical property of lacquer wax were basically consistent with those reported in literature,and the composition mainly included long chain fatty acids,such as palmitic acid,oleic acid and stearic acid.The experiment has proved that lacquer wax extracted by ultrasound-assisted enzymatic method could provide a reference for the resources comprehensive utilization of lacquer seed.
