白杨素与咖啡酸苯乙酯抗肿瘤活性比较

摘要: 研究白杨素和咖啡酸苯乙酯对不同肿瘤细胞的细胞毒性。正常培养的乳腺癌细胞(MCF-7、MDA-MB-231)、肺腺癌细胞(A549)、宫颈癌细胞(Hela)及血管内皮细胞(HUVECs)经不同浓度的白杨素和咖啡酸苯乙酯(20、40、80、160 μM)分别处理24和48 h,倒置显微镜观察细胞形态,SRB法检测细胞存活率,吖啶橙染色和Hoechst 33258检测了对MDA-MB-231细胞凋亡的影响;划痕法检测了对细胞迁移的影响;荧光探针DCFH-DA、JC-1检测细胞内活性氧(ROS)和线粒体膜电位,免疫细胞化学法检测了NF-κB p65水平。结果表明,白杨素和咖啡酸苯乙酯以时间和剂量依赖的方式抑制肿瘤细胞增殖和迁移,上调MDA-MB-231细胞内的ROS,下调细胞内的线粒体膜电位和NF-κB p65水平。白杨素和咖啡酸苯乙酯是潜在的抗肿瘤活性的天然产物。

Abstract: To study the anti-tumor activities of chrysin and caffeic acid phenethyl ester (CAPE).The normal cultured human breast cancer (MCF-7,MDA-MB-231),human cerrical cancer cells (HeLa),human lung carcinoma (A549) and human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of chrysin and caffeic acid phenethyl ester (20,40,80,160 μM) for 24 and 48 h,respectively.The cell viability was measured by SRB method.Hoechst 33258 staining and acridine orange staining were used to observe apoptotic morphology and nuclear fragmentation,respectively.The cell migration was detected by scratch method.Intracellular reactive oxygen species (ROS) level and mitochondria memberane potential were determined by utilizing a fluorescent probe,2′,7′-dichlorodihydrofluorescin (DCHF) and JC-1,and the level of NF-κB p65 was tested by immunofluorescence.The results showed that chrysin and CAPE inhibited different tumor cells proliferation and migration,and upregulated ROS level and downregulated mitochondrial membrane potential and NF-κB p65 levels.Chrysin and caffeic acid phenethyl ester were potential anti-tumor agents.
