中药调控阿尔茨海默病tau蛋白过度磷酸化的研究进展

摘要:

阿尔茨海默病(Alzheimer's disease,AD)是神经系统退行性疾病,常伴有情感障碍、生活学习能力降低等症状,其发病不可逆,且进行性加重,已成为严重危害老年人健康的全球性公共卫生事件。tau蛋白过度磷酸化是导致AD的重要节点,tau蛋白磷酸化水平受激酶和磷酸酶的双重调节,既往研究表明中药可多途径调控tau蛋白磷酸化水平并有防治AD的作用。本文从中药对tau蛋白激酶的调控、磷酸酶的调控、磷酸酶和激酶的共同调控3方面进行综述,为研发有效防治AD的中药新药提供思路和参考。

Abstract:

Alzheimer's disease (AD) is degenerative disease of the nerve system,often accompanied by symptoms such as mood disorder,living and learning ability reduction,and others.As such symptoms are irreversibly worsening,this disease has become a global public health problem that seriously threatens the health of the elderly.Based on the author’s understanding that the tau protein hyperphosphorylation is a key factor leading to AD and that the tau protein phosphatase level and the tau protein phosphorylation may be regulated in multiple ways by traditional Chinese medicine (TCM) in the treatment of AD,as proven by finding already made,this article is an attempt to present new ideas and approaches in TCM pharmacy and TCM treatment of AD,in the light of tau protein kinase regulation,phosphatase regulation and kinase and phosphatase dual regulation.
