红雪茶提取物的抗氧化活性及其机理初探

摘要:

以红雪茶为试验材料,采用人类肝癌细胞(HepG2)和秀丽线虫(Canorhabditis elegans)为模型研究红雪茶醇提物(Lethariella spp.ethanol extract,LEE)的抗氧化能力及其可能的作用机制。结果表明:400~800 μg/mL LEE能够有效提高丙烯酰胺诱导的氧化损伤下的HepG2细胞活力,降低细胞的凋亡。在体内环境下,LEE具有优秀的抗氧化胁迫的保护潜力。0.01 mg/mL LEE对线虫的生长发育以及生殖不产生影响。在热应激条件下,经LEE预处理后野生型N2线虫的平均寿命由10.00 ± 0.00 h延长至12.33 ± 0.58 h,延长了23.33%,但LEE不能延长热应激环境下age-1daf-2daf-16突变体线虫的寿命。综上所述LEE具有显著的体内外抗氧化生物活性,其作用依赖于age-1daf-2daf-16三个基因,表明LEE的抗氧化胁迫的保护潜力的机制可能依赖于胰岛素信号通路。

Abstract:

In this study,HepG2 cells and Caenorhabditis elegans were used as models to study the antioxidant capacity of Lethariella spp. ethanol extract (LEE) and its possible mechanism of action.The results showed that 400-800 μg/mL LEE can effectively increase the cell viability of HepG2 cells and reduce cell apoptosis on the oxidative stress induced by acrylamide.In the in vivo environment,LEE exhibited an excellent potential protection on C. elegans against oxidative stress.0.01 mg/mL LEE showed no negative effect on the growth and reproduction of nematodes.Under thermal stress,the mean survial of wild-type N2 nematodes pretreated with LEE was prolonged by 23.33% from 10.00 ± 0.00 h to 12.33 ± 0.58 h.However,LEE failed to extend the survivals of age-1,daf-2 and daf-16 mutant nematodes under heat stress.In summary,LEE possesses a promising anti-oxidant biological activity in vivo and in vitro,and its mechanism of action may depend on the three genes of age-1,daf-2 and daf-16  in the insulin signaling pathway.
