红腺忍冬山银花绿原酸的分离纯化研究

摘要: 以红腺忍冬山银花叶为原料,用70%乙醇加热回流提取绿原酸,提取液经大孔树脂树脂分离纯化,通过树脂筛选,最佳树脂为D101大孔树脂,利用大孔树脂D101对红腺忍冬山银花叶中绿原酸的分离纯化工艺进行了优化研究。实验结果表明:最佳上柱条件为:固液比6∶〖KG-*2〗1(mL∶〖KG-*2〗g)、上样液pH为2、流速60 滴/min;最佳洗脱工艺条件为:洗脱溶液为20%乙醇,洗脱液体积30 mL(6倍树脂床体积),流速40 滴/min,洗脱溶液pH为7。在最佳工艺条件下,红腺忍冬叶提取液经大孔树脂分离纯化后绿原酸平均收率为1.41%,绿原酸平均纯度为458%,该分离纯化方法具有操作简单、生产周期短和成本低等优点,适用于红腺忍冬山银花绿原酸的分离纯化及规模化工业生产。

Abstract: Chlorogenic acid was extracted from Lonicera hypoglauca Miq leaf with ethanolheat reflux methods and separated and purified by macroporous resin method,D101 is as the optimum macroporous resin by screening.studies was made on separation and purification of chiorogenic acid from Lonicera hypoglauca Miq leaf extraction by macroporous resin for screening the optimal conditions.UV maximum absorption of chlorogenic acid is used as an indicator to test wavelength at 320nm.The optimum adsorption technology is as follows:resinliquid ratio as 1 g:6 mL,adsorption solution pH 2,adsorption solution flow rate of 60/min .The optimum elution technology is as follows:elution solution as 20% ethanol,volume of elution solution is 30 mL, the flow rate was 40/min, elution solution pH7.Under optimal conditions of separation and purification, average recovery rate and purity of chlorogenic acid from Lonicera hypoglauca Miq leaf extraction is 1.403% and 45.77% respectively.The method developed in the paper has the advantages of simple operation,short production cycle and low cost which appropriate for the separation and purification of chiorogenic acid from Lonicera hypoglauca Miq leaf extraction and industry scale production.
