Effects of four Chinese herbs for lung channel tropism on energy metabolism in mice model with lung heat syndrome based on AMPK/PGC-1α signaling pathway

摘要:

研究陈皮(Citri Reticulatae Pericarpium,CP)、白芷(Angelicae Dahuricae Radix,AR)、葶苈子(Descurainiae Semen,DS)、射干(Belamcandae Rhizoma,BR)四味归肺经中药对肺热证模型小鼠能量代谢的影响及机制。建立肺热证小鼠模型进行CP、AR、DS和BR干预,通过苏木精-伊红染色法观察肺组织形态学变化,酶联免疫吸附法检测血清中促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)、多巴胺β羟化酶(dopamine beta hydroxylase,DBH)含量,生化试剂盒检测小鼠肝组织中Na-K-ATP酶、琥珀酸脱氢酶(succinate dehydrogenase,SDH)、乳酸脱氢酶(lactate dehydrogenase,LDH)和超氧化物歧化酶(superoxide dismutase,SOD)水平,免疫组织化学法及实时荧光定量聚合酶链式反应检测肝组织腺苷酸活化蛋白激酶(adenosine monophosphate-activated protein kinase,AMPK)、过氧化物酶体增殖物激活受体γ共激活因子-1α(peroxisome proliferator-activated receptor γ coactivator-1α,PGC-1α)蛋白及mRNA表达水平。结果表明与模型组相比,CP、AR、DS和BR不同程度降低血清中ACTH、DBH及肝组织中Na-K-ATP酶、SDH和SOD表达水平以及AMPK、PGC-1α蛋白及mRNA表达水平,增强LDH表达水平。CP、AR、DS和BR可能通过抑制AMPK/PGC-1α信号通路调控肺热证模型小鼠能量代谢。

Abstract:

The aim of this study is to investigate the effects of Citri Reticulatae Pericarpium (CP),Angelicae Dahuricae Radix (AR),Descurainiae Semen (DS) and Belamcandae Rhizoma (BR) on energy metabolism in mice model with lung heat syndrome and its mechanism.The mouse model with lung heat syndrome was established and treated with CP,AR,DS and BR.The morphological changes of lung were observed by hematoxylin-eosin staining method,and the contents of adrenocorticotropin (ACTH) and dopamine-β hydroxylase (DBH) in serum were detected by enzyme-linked immunosorbent assay method.The levels of Na+-K+-ATPase,succinate dehydrogenase (SDH),lactate dehydrogenase (LDH) and superoxide dismutase (SOD) in mouse liver tissue were detected by biochemical kit.The protein and mRNA expression levels of adenylate activated protein kinase (AMPK) and peroxisome proliferator-activated receptor-γ coactivator-1α(PGC-1α) in liver tissue were detected by immunohistochemistry and real-time quantitative polymerasechain reaction.The results showed that compared with the model group,CP,AR,DS and BR reduced the expression levels of ACTH,DBH in serum and Na+-K+-ATPase,SDH,SOD in liver tissue,as well as the expression levels of AMPK,PGC-1α protein and mRNA in liver tissue,and enhanced the expression level of LDH.CP,AR,DS and BR may regulate the energy metabolism of lung heat syndrome model mice by inhibiting the AMPK/PGC-1α signaling pathway.
