Extraction of β-Carotene from Spirulina Using Ethanol and Acetone-Ammonium Sulphate Aqueous Two-Phase System

Abstract: In this study,an aqueous two-phase system composed of 15% (W/W) ethanol and acetone (v/v=1:2) 24% (W/W) ammonium sulphate was selected to extract β-carotene from Spirulina. The effect of crude extract concentration,pH,extraction temperature on the extraction efficiency of  β-carotene were investigated by single factor and Box-Behnken design methods.The results showed that the optimal conditions for the extraction of β-carotene were:crude extract concentration of 6% (W/W),pH of 8.0 and extraction temperature of 30 ℃.The maximal extraction efficiency (94.55%) was obtained under the optimal conditions.These results suggested that ATPE is efficient in extracting β-carotene and has the potential to be used in natural β-carotene extraction industry.
