Effect of Crude Polysaccharide from Phyllanthus emblic Fruit on Oxidative Stress in HepG-2 Cells

Abstract: In order to study the effects of crude polysaccharide from Phyllanthus emblic fruit on the growth of HepG-2 cells and oxidative stress in HepG-2 cells. Hepatoma carcinoma cells HepG-2 were treated by a crude emblic polysaccharide (CEPS) in different concentrations for 24,48,72 h.Then cellular morphology was observed using inverted biological microscope,cellular proliferation inhibition rate was detected by MTT colorimetric method and superoxide dismutase (SOD) activity,lactic dehydrogenase (LDH) level,glutathion peroxidase (GSH-Px) level,malonadialdehyde (MDA) content were also determined.The results showed that with the increasing of CEPS concentration,the survival rate of HepG-2 cells was decreased,adherent growth became anomaly,dead cells increased and the content of MDA,SOD,LDH,GSH-Px appeared increasing trend.Preliminary judging that HepG-2 cells were injured under the treatment of ECPS,leading to peroxide MDA content increased,and in order to protect themselves,the corresponding antioxidant enzymes increased their content.Indicating that emblic crude polysaccharide affected oxidative stress in HepG-2 cells to induce injury of HepG-2 cells and decrease proliferative activity of HepG-2 cells.
