Effects of Trillin on Hydrogen Peroxide-induced Oxidative Stress Injury and Inflammatory Factor in PC12 Cells

Abstract: To investigate effects of trillin on hydrogen peroxide(H2O2)-induced oxidative stress injury and inflammatory factor expression in PC12 cells.MTT and LDH assay were used to determine the effects of trillin on PC12 cell model,respectively.Reactive oxygen species(ROS) and malondialdehyde(MDA) contents,superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) were detected by according kits,respectively.Furthermore,Sirt1,NF-κB and TNF-α protein expressions were determine by Western blot assay.Trillin(5-20 μM) significantly improved H2O2-induced PC12 cell viability and anti-oxidant enzyme activity.Moreover,trillin up-regulated Sirt1 protein expression,and down-regulated NF-κB and TNF-α protein expression.Its protective effect was possibly through enhancing anti-oxidant capability,decreasing inflammatory factor and regulating Sirt1/NF-κB signaling pathway.
