Protective Effects of Peripheral Blood of Total Saponins of Sanguisorba officinalis L.on Myelosuppressive Mice induced by 60Co-γ ray and Its Associated Mechanism

Abstract: Protective effects and mechanism of total saponins of Sanguisorba officinalis L.(TSSO) on myelosuppressivemiceinduced by 60Co-γ ray was investigated.The myelosuppressivemice modelwas established by irradiating the whole body one time with 650 cGy 60Co-γ  ray.Then the myelosuppressive mice were treated with TSSO(1,0.5 and 0.1 mg/kg,i.g.) once a day,for total 10 days.The levels of WBC,RBC,HCT,PLT and PCT were measured at the 4th, 7th and 10th day.The next day,the liver,spleen and thymus was collected for counting organ index.The numbers of bone marrow cells were also counted and the mRNA expressions of GM-CSF,IL-3,SCF and c-Kit were measured by qPCR analysis.The results showed that TSSO can improve the levels of WBC,RBC,HCT,PLT,PCT,BMC,and the mRNA expression of GM-CSF,IL-3,SCF,and c-Kit in TSSO groups significantly.Thus,TSSO can alleviate myelosuppression induced by 60Co-γ ray. The improvement of the mRNA expressions of GM-CSF,IL-3,SCF and c-Kit may be the mechanisms.
