Extraction and Antitumor Activity of Active Ingredients in Bovistella radicata

Abstract: Bovistella radicata was used as raw material and petroleum ether was used as solvent to carry out Soxhlet extraction,and the petroleum ether extract was obtained after drying.After drying the residue,the same method was followed by adding ethyl acetate,n-butanol,ethanol,distilled water to afford ethyl acetate extract,n-butanol extract,ethanol extract and water extract.The inhibitory effects of different solvent extracts on the cell viability of MDA-MB-231 and Hela cells were detected by MTT assay.The intracellular reactive oxygen species (ROS),mitochondrial membrane potential detection and DAPI detection of apoptosis in MDA-MB-231 cells treated with n-butanol extract were observed by inverted fluorescence microscopy.The results showed that the extracts of B.radicata can decrease the viability of MDA-MB-231 and Hela cells,especially the n-butanol extract.When the concentration of n-butanol extract was 200 μg/mL,the cell viability was significantly inhibited.Compared to Hela,MDA-MB-21 was more sensitive to the effect of the extracts.When treated with the n-butanol extract,the activity of MDA-MB-231 cells decreased significantly,the mitochondrial membrane potential decreased significantly,and the intracellular reactive oxygen species increased significantly,and the mitochondrial pathway was induced by ROS.
