 Effects of Qi-Tonifying Decoction on TGF-β1/Smad Signaling Pathway in Rats with Hepatic Fibrosis Induced by Carbon Tetrachloride

Abstract: To investigate the therapeutic effect and mechanism of Qi-Tonifying Decoction (Astragalus membranaceus and Atractylodes macrocephala) on hepatic fibrosis induced by CCL4 in rats.Hepatic fibrosis model was established by intraperitoneal injection of 40% carbon tetrachloride (CCL4) corn oil solution combined with high fat and low protein diet.The drugs were given to each group for 12 weeks.HE and Masson staining were used to observe the morphological changes of hepatic tissues.Biochemical analysis was used to detect liver function in serum.Enzyme-linked immunosorbent assay(ELISA) was used to detect collagen content in serum.The expression of TGF-beta 1,T beta R I,Smad3,Samd7,alpha -SMA and HGF in liver tissue was detected by RT-qPCR.After administration,Qi-Tonifying decoction and colchicine could reduce collagen deposition and improve liver function in rats,and its mechanism is related to inhibiting TGF-beta 1/Smad signaling pathway.
