Network pharmacological study on the potential mechanism of Danggui Buxue Decoction in the treatment of diabetic nephropathy

Abstract:

To study the potential material basis and mechanism of Danggui Buxue Decoction in the treatment of diabetic nephropathy (DN).Firstly,28 active components and 188 DN-related targets in Danggui Buxue Decoction were screened by TCMSP database,and then the drug-compound-target network diagram was constructed by Cytoscape software.The key targets of Danggui Buxue Decoction in the treatment of DN were analyzed by CentiScaPe plug-in,and 12 significant effective compounds and 13 key targets were determined by network topology analysis.Furthermore,the GO enrichment analysis and KEGG pathway enrichment analysis were carried out on the key targets by using the ClusterProfiler package of R software.It was found that the active components such as quercetin,kaempferol and formononetin in Danggui Buxue Decoction could regulate 162 related pathways such as AGE-RAGE,IL-17 and HIF-1 by PTGS2,GABRA1 and ncoa2,involving cytokine receptor binding,cytokine activity,phosphatase binding,serine hydrolase activity,antioxidant activity and kinase regulatory activity.There are 131 related biological processes,which play a synergistic role in the treatment of DN.
