Prediction of the effects of total flavonoids of Kunlun chrysanthemum in the treatment of non-small cell lung cancer by network pharmacology and its preliminary verification study

Abstract:

The composition analysis of the total flavonoids of Kunlun chrysanthemums was carried out by the high performance liquid chromatography(HPLC).The “component-target-pathway” network was constructed through the method of network pharmacology.Based on the prediction results,the CCK8 assay was used to detect the proliferation inhibitory activity of Kunlun chrysanthemum total flavonoids(KCTF)at different concentrations on A549 cells,flow cytometry was used to detect cell apoptosis,and a cell scratch assay was used to observe the effects of KCTF on migration ability of A549 cells.RT-qPCR was used to detect the expression of Bax,Bcl-2,and MMP-2 genes,and Western blot was used to detect the expression of related apoptotic proteins Bax and Bcl-2.The results showed that 68 KCTF components were obtained by HPLC analysis,27 active components were obtained after screening,and 217 KCTF intervention non-small cell lung cancer(NSCLC) related targets were constructed through the relevant database platform.The results of KEGG pathway enrichment analysis showed that the key nodes of KCTF intervention in NSCLC were mainly enriched into 76 signaling pathways such as cell proliferation,apoptosis and cycle regulation,viral carcinogenesis,and other tumor pathways.The results of cell-level verification experiments showed that KCTF significantly inhibited the proliferation of A549 cells,promoted cell apoptosis,and inhibited cell migration;RT-qPCR experiments verified that KCTF up-regulated the expression of Bax and down-regulated the expression of Bcl-2 and MMP-2 genes;Western blot results showed that KCTF significantly up-regulates the expression of Bax/Bcl-2,and induces cell apoptosis.In summary,KCTF can intervene in NSCLC through multiple targets and multiple pathways,and its anti-tumor activity may be related to affecting the expression of apoptosis-related genes Bax and Bcl-2.
