Effect of piceatannol on proliferation,migration and invasion of endometrial cancer HEC-1A cells

Abstract:

The purpose of this study was to investigate the preliminary antitumor effects of piceatannol (PIC) on endometrial cancer cells HEC-1A.The effects of different concentrations of PIC on the proliferation,apoptosis,migration,and invasion abilities of HEC-1A cells was detected by CCK-8 test,plate cloning experiment,flow cytometry,wound healing assay,and Transwell assay.Western blot was used to detect the expression level of cell proliferation related proteins and invasion migration related proteins.The results showed that compared with the control group,PIC significantly reduced the migratory distance and number of invading cells (P<0.01),inhibited the proliferation and clonogenic ability of HEC-1A cells,induced apotosis (P<0.01),and down-regulated the expression levels of p-Akt,p-Erk1/2,p-p38MAPK,β-catenin,CD44 and Slug proteins,and up-regulated the expression levels of E-cadherin protein (P<0.01).In conclusion,PIC demonstrated concentration-dependent suppression of endometrial cancer HEC-1A cell proliferation,migration and invasion.The mechanism may be connected to the inhibition of the AKT/ERK/MAPK signaling pathway and impacting the Wnt/β-catenin signaling pathway and alterations in epithelial-mesenchymal transition markers.
