Seperation and Identification of a Novel Halophile

Abstract:

A marine bacterium, designated strain S3-22, was isolated from the ballast water sample from Zhoushan archipelago, China. 16S rDNA sequences similarity of strain S3-22 to all recognized bacterial species was not greater than 97 %. The DNA G+C content was 54.9 mol %. The dominant fatty acid was iso-C17:1ω 9c (24.99 %). The major respiratory quinine was MK-5. Cells of strain S3-22 were gram-negative. Optimal growth of strain S3-22 was at pH 7.0, 30–37 ℃ and 3% (w/v) NaCl. It was halophilic and tested positive for catalase, oxidase, amylase, and esterase. In addition, it could deoxidize the nitrate. Based on the evidence of 16S rDNA sequences similarities, phylogenetic analysis and cellular and molecular level identification, the halophile formed a phyletic lineage which was distinct from the known Kordiimonas species. Strain S3-22 represented a novel species in the Kordiimonas, its Genebank Accession number was FJ847942.
