Simultaneous Determination of Four Active Components in Tibetan Herb Gentiana purdomii Marq. by HPLC

Abstract: A quantitative method of the simultaneous determination of swertiamain,gentiopicroside,sweroside and Isoorientin in Gentiana purdomii Marq.was established using RP-HPLC.The results showed that the contents of swertiamain,gentiopicroside,sweroside and Isoorientin were 0.108%,1.46%,2.21% and 1.61%, respectively. The HPLC analysis conditions were as follows:XDB-C18 column (250 mm × 4.6 mm,5 μm) was used for chromatographic separation and methanol0.01% lacetic acid were used as the mobile phases with gradient elution;The flow rate was 1.0 mL/min with a column temperature at 25 ℃,and the UV detection wavelength was set at 260 nm.The assay demonstrated that the developed method had adequate accuracy and selectivity.It was suitable for the determination of four active components in G.purdomii. Hence,it can provide additional evidence for the quality evaluation of G.purdomii.
