养阴解毒汤治疗新冠肺炎的临床疗效及其作用机制的网络药理学研究

摘要:

探讨养阴解毒汤对新型冠状病毒肺炎(COVID-19)的临床疗效及其作用机制。随机选取2020年1月28日到2020年2月28日贵州省将军山医院收治的COVID-19患者作为研究对象,观察养阴解毒汤联合常规治疗前后患者的一般情况、症状体征、肝肾功能指标、核酸转阴情况、CT影像学的变化等情况,同时运用网络药理学及分子对接分析养阴解毒汤治疗COVID-19的潜在作用靶点、活性成分及作用机制。养阴解毒汤能改善新型冠状病毒肺炎患者的临床症状,应用网络药理学分析养阴解毒汤治疗COVID-19的作用靶点,得到潜在作用靶点14个,GO功能富集56条目,KEGG通路富集3条,分子对接结果表明山奈酚、β-谷甾醇、印度榅桲素与受体蛋白SARS-CoV-2 3CL水解酶形成构象能量低,结构稳定,结合活性高。养阴解毒汤具有治疗COVID-19的效果,其作用可能与山奈酚、β-谷甾醇、印度榅桲素等化合物从各靶点发挥抗病毒、抗炎、抗氧化等作用有关。

Abstract:

To explore the clinical effect and mechanism of Yangyin Jiedu Decoction on coronavirus disease 2019(COVID-19).The patients with COVID-19 who were treated in general hospital of Guizhou province from January 28,2020,to February 28,2020,were randomly selected as the study object.Their general situation,symptom and signs,liver and kidney function index,nucleic acid Yin conversion and CT imaging changes,before and after the treatment of Yangyin Jiedu Decoction combined routine treatment,were observed.Meanwhile,the network pharmacology and molecular docking was used to analyze the potential targets,components and mechanism of action.Yangyin Jiedu Decoction can improve the clinical symptoms of patients with COVID-19,the network pharmacology analysis showed that 14 potential targets were obtained,56 items were enriched by GO function,and 3 pathways were enriched by KEGG analysis.The results of molecular docking showed that kaempferol,β-sitosterol and marmesin had low conformation energy,stable structure and high binding activity when bind to the receptor protein SARS-CoV-2 3CL hydrolase.Yangyin Jiedu Decoction has a therapeutic effect on COVID-19,which may be related to the antiviral,anti-inflammatory and anti-oxidation effects of kaempferol,β-sitosterol,marmesin and other compounds from various targets.
