蝇蛆酶解产物体外抗氧化活性研究

In vitro antioxidant activity of hydrolysates of fly maggots

本研究以鲜蝇蛆为原料,采用两种蛋白酶水解制备出酶解产物,研究了酶解产物的体外抗氧化活性;采用凝胶过滤色谱检测酶解产物的多肽分子量分布,用氨基酸自动分析仪测定其氨基酸组成和含量;最后用超滤膜初步分离出分子量在3 000 Da以下的小分子肽组分,并测定了其抗氧化活性。结果表明,用中性蛋白酶和碱性蛋白酶水解蝇蛆获得的酶解产物均具有良好的自由基清除活性和Fe2+螯合能力。两种酶解产物中分子量在3 000 Da以下的小分子肽均达到97%以上。蝇蛆酶解产物主要包含17种氨基酸,中性蛋白酶和碱性蛋白酶酶解产物与抗氧化相关的3类氨基酸(疏水性氨基酸、酸性氨基酸、碱性氨基酸)分别占83.48%和83.84%。经超滤膜初步分离后获得的分子量小于3 000 Da的小分子肽的抗氧化活性显著提高,两种酶解产物3 000 Da以下的小分子肽组分对DPPH自由基、ABTS自由基、羟基自由基及Fe2+螯合的IC50分别为0.40 mg/mL和0.26 mg/mL、0.35 mg/mL和0.33 mg/mL、0.92 mg/mL和1.00 mg/mL及1.09 mg/mL和1.75 mg/mL。综上,利用中性蛋白酶和碱性蛋白酶制备的蝇蛆小分子肽具有较强的抗氧化能力,氨基酸种类丰富、营养全面,为天然抗氧化药物的开发和应用提供后备资源和理论基础。

Abstract:

In this study, fresh fly maggots were used as raw materials, two kinds of proteases were used to prepare the protein hydrolysates, and the in vitro antioxidant activity of hydrolysates were investigated. The molecular weight distribution of peptides of hydrolysates were detected by gel filtration chromatography, and the amino acid composition and content were determined by automatic amino acid analyzer. Finally, the small peptide with molecular weights of less than 3 000 Da were preliminarily separated by ultrafiltration membranes and the antioxidant activities of the peptides were determined. The results showed that neutral protease hydrolysate and alcalase hydrolysate obtained by fly maggots both had good free radical scavenging activity and Fe2+ chelating ability. The fractions with molecular weights below 3 000 Da reached more than 97% for both hydrolysates.The hydrolysate mainly contained 17 amino acids, and the three classes of amino acids (hydrophobic amino acid, acidic amino acid, basic amino acid) associated with antioxidants in the neutral protease hydrolysate and alcalase hydrolysate were accounted for 83.48% and 83.84%, respectively. The antioxidant activities of small peptides with molecular weights less than 3 000 Da obtained after preliminary separation by ultrafiltration membranes were significantly improved, and the IC50 of the small peptide with hydrolysates below 3 000 Da against DPPH radical, ABTS radical, hydroxyl radical, and Fe2+ chelating were 0.40 mg/mL and 0.26 mg/mL, 0.35 mg/mL and 0.33 mg/mL, 0.92 mg/mL and 1.00 mg/mL, 1.09 mg/mL and 1.75 mg/mL, respectively. In conclusion, both neutral protease hydrolysate and alcalase hydrolysate prepared from fly maggots has strong antioxidant capacity, rich amino acids and high nutrition, which can provide reserve resources and theoretical basis for the development and application of natural antioxidant medicines.
