桑叶酚类物质在体外模拟消化过程中生物可接受率和抗氧化活性的研究

摘要:

为探究桑叶酚类物质在体外模拟消化过程中的释放量、生物可接受率和抗氧化活性的变化规律,以果桑和叶桑为原料,采用体外模拟消化模型,通过分光光度法和HPLC法测定其酚类物质含量。结果表明,桑叶多酚、黄酮、芦丁和绿原酸在消化过程中逐渐降低,且肠消化阶段减少量最大,而槲皮素在胃消化阶段的释放量较消化前升高,但经肠消化后显著降低。模拟消化前,桑叶结合态酚的抗氧化活性明显高于游离态酚。在体外模拟消化过程中,桑叶酚类物质的抗氧化能力显著下降(P<0.05)。经过胃肠消化后,果桑多酚与叶桑槲皮素生物可接受率最高,酚类物质的生物接受率均小于1。通过相关性分析发现,桑叶的抗氧化活性与释放的黄酮、多酚、芦丁以及绿原酸释放量之间存在显著的正相关性。综上所述,体外模拟消化会对桑叶酚类物质造成影响,研究结果为进一步揭示桑叶中酚类物质在体内消化利用规律提供一定的理论参考。

Abstract:

To explore the changes in the release of phenolic substance,bioaccessibility,and antioxidant activity in fruit mulberry and leaf mulberry leaves during in vitro simulated digestion,the contents of phenolic compounds in mulberry leaves were determined by HPLC and spectrophotometry.The results revealed a gradual decrease in polyphenols,flavonoids,rutin,and chlorogenic acid in mulberry leaves during digestion,with the most significant reduction occurring during intestinal digestion.Quercetin release increased during gastric digestion compared with that before digestion,but decreased significantly after intestinal digestion.Prior to in vitro simulated digestion,bound phenols in mulberry leaves exhibited significantly higher antioxidant activity than free phenols.During the simulation digestion process,the antioxidant capacity of phenolic compounds in mulberry leaves decreased significantly (P<0.05).Following gastric and intestinal digestion stages,bioaccessibility was highest for polyphenols in fruit mulberry and quercetin in leaf mulberry,however it remained less than 1 for all phenolic compounds analyzed.Correlation analysis indicated a positive correlation between antioxidant activity of mulberry leaves and the release levels of flavonoids,polyphenols,rutin,and chlorogenic acid.In conclusion, in vitro simulated digestion affects phenolic compounds in mulberry leaves.This research provides a theoretical basis for further understanding the utilization patterns of phenolic compounds in mulberry leaves during in vivo digestion.
