三种卷柏属植物对人单核细胞白血病U937细胞的增殖抑制作用

Anti-proliferative Effect of Three Selaginella Plants on Human Monocytic Leukemia U937 Cell Line

摘要: 为了观察中华卷柏、鹿角卷柏、卷柏等三种卷柏属植物的甲醇总提物、乙酸乙酯部位、石油醚部位样品对体外培养的人单核细胞白血病细胞U937细胞的增殖抑制作用。通过常规传代培养人单核白血病U937细胞,再用噻唑兰比色法(MTT)检测不同浓度样品作用于U937细胞12、24 h和48 h后的细胞增殖情况。中华卷柏和卷柏对U937细胞的增值抑制呈时间和剂量依赖性。鹿角卷柏对U937细胞的增值抑制呈剂量依赖性。药物作用24 h时,三种卷柏属植物对U937细胞均有显著抑制作用,其半数抑制浓度(IC50)分别为中华卷柏甲醇总提物102.54 μg/mL、乙酸乙酯部位87.95 μg/mL、石油醚部位91.63 μg/mL;鹿角卷柏甲醇总提物118.69 μg/mL、乙酸乙酯部位126.03 μg/mL、石油醚部位135.8 μg/mL;卷柏甲醇总提物103.92 μg/mL、乙酸乙酯174.75 μg/mL、石油醚102.64 μg/mL。证明了三种卷柏均能有效抑制U937细胞的增殖,为后续对卷柏属植物抗癌机制的研究提供了方向。

Abstract: To investigate the anti-proliferative effect of methanol extracts,ethyl acetate fraction,petroleum ether fraction extracted from Selaginella tamariscina (Beauv.) Spring, Selaginella rosii (Bak.) Warb, Selaginella sinensis (Desv.) Spring on human monocytic leukemia U937 cell line. U937 cells were cultivated by conventional way. MTT assay was used to measure the proliferation of U937 cells after treatment with different concentrations of methanol extract and different polar fractions from the Selaginella plant. S.tamariscina and S.sinensis caused the tumor cells death in a dose-dependent manner and time-dependent manner, S.rosii caused the tumor cells death in a dose-dependent manner.The methanol extracts,ethyl acetate fraction,petroleum ether fraction extracted from S.tamariscina, S.rosii, S.Sinensis can significantly inhibit the proliferation of U937 cells at IC50 of 102.54,87.95,91.63 μg/mL;118.69 μg/mL,135.8 μg/mL,95.11 μg/mL;103.92 μg/mL,174.75 μg/mL,102.64 μg/mL,respectively.In conclusion,the three Selaginella plants can significantly inhibit the proliferation of U937 cells.
