Triterpene Glycoside Des-echinoside A Inhibits Human Lung Cancer Cell Line 95D Metastasis via NF-κB Signaling Pathway

Abstract: A triterpene glycoside Des-echinoside A was separated by high speed counter current chromatography from sea cucumber Pearsonothuria graeffei. The effects and molecular mechanism of Des-echinoside A on tumor metastasis in high-metastatic (95D) human lung cancer cells were evaluated.The results showed that Des-echinoside A could inhibit 95D cell proliferation (P<0.01),migration (P<0.01),invasion (P<0.01),adhesion(P<0.01) and decrease the blood vessel growth on the chick embryo chorioallantoic membrane(CAM) remarkably.Des-echinoside A could also reduce the mRNA expression of uPA,MMP-2/-9,VEGF,NF-κB (P<0.05,P<0.01),and promote the mRNA expression of TIMP-1/-2(P<0.01).It is suggested that Des-echinoside A could prevent the metastasis through specific inhibition of NF-κB signaling pathway.
