Abstract: To explore the intervention effect of Huayu Tongluo Traditional Chinese Medicine on nuclear factor kappa B in renal cortex of blood stasis syndrome rats with diabetic nephropathy (DN).Ten rats were selected as normal group from sixty rats,the models were established by high-fat diet combined with low-dose streptozotocin intraperitoneal injection,and then were randomly divided into model group,irbesartan treatment group and Chinese medicine group after successful modeling.Rats in each treatment group were fed with corresponding drug intragastrically.At the end of the 16th week,water intake,food intake,urine output,body weight and fasting blood glucose were detected.24 h urinary total protein (24 hUTP) of rats were measured at different time point (3rd day,8th week and 16th week) in each group.Renal cortex were conserved to observe the pathological injury and the mRNA and protein expression of NF-κB p65 in renal cortex were detected by Real-time PCR,Western-blot.The results showed that compared with normal group,model group rats appeared obvious polydipsia,polyphagia,polyuria,emaciation (P<0.05),8th and 16th week of 24 hUTP increased significantly (P<0.05),the mRNA and protein expression of NFκB p65 enhanced (P<0.05).Renal pathology appeared obvious injury.Compared with model group,24 hUTP of two intervention groupsdecreased significantly at 8th week and 16th week (P<0.05),the mRNA and protein expression of NFκB p65 decreased significantly (P<0.05),water intake,urine output,body weight of rats in Chinese medicine group improved significantly (P<0.05).Compared with irbesartan group,water intake,24 hUTP at 16th week of the Chinese medicine group and the protein expression of NF-κB p65 decreased significantly (P<0.05).The results showed that inhibiting the high expression of NF-κB p65 can be one of the targets of the medicine on the protective effect of the kidney of DN rats.
