Hypolipidemic Effects and Mechaniasm of Tea Plant New Strain Camellia sinensis CV.CFT-1 Extract

Abstract: The aim of this study was to investigate the hypolipidemic effect and mechanism of tea plant new strain Camellia sinensis CV.CFT-1 extract (CFT-1).Forty rats were randomly divided into 4 groups,the normal group (CON),the hyperlipidaemia model group (HFD),the CFT-1 green tea group (CFT-1) and the ordinary green tea group (Fuyun6).Hyperlipidaemia rats model were established by feeding rats with a high-fat diet.Green tea extracts of CFT-1 or Fuyun6 was administered once a day for eight weeks by gavage as pretreatment at 200 mg/kg.The blood lipid levels,the activities of ALT and AST in serum,antioxidant activity in serum and liver were analyzed.The levels of tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6) and interleukin-10 (IL-10) were analyzed by ELISA.Thepathological changes of liver tissuewere observed by hematoxylin and eosin staining.The protein and mRNA expression of fatty acid synthetase(FAS),low density lipoprotein receptor(LDLR),acetyl-coa carboxylase (ACC),sterol regulatory element binding protein-1c (SREBP-1c),CCAAT/enhancer binding protein α (C/EBPα),Peroxidase proliferation activated receptor-a (PPARα),carnitine palmitoyltransferase-1 alpha (CPT-1α) and 1-amino-cyclopropane-1-carboxylate oxidase (ACO)genes related to lipid metabolism and anti-oxidant were assayed by Western blot and qRT-PCR.Green tea extract of CFT-1 significantly decreased the weight gains,levels of serum TC,TG,LDL-C induced by high-fat diet and significantly increased the level of serum HDL-C.Histologically,the extract reduced the formation of lipid droplets,alleviated the degree of fatty degeneration and prevented liver fibrosis in livers.The activities of SOD,GSH-Px and CAT were also significantly increased in serum and liver of rats treated with the extract,while the content of MDA was decreased.The pretreatment with green tea extract of CFT-1 significantly decreased levels of serum ALT,AST,IL-6,TNF-ɑand increased levels of serum IL-10,decreased the protein and mRNA expression degrees of ACC,FAS,SREBP-1c and C/EBP-α,while up-regulated the protein and mRNA expression degrees of LDLR,CPT-1α,PPAR-α,and ACO in liver of hyperlipidemia rats.Moreover,these results reveals the green tea extract of CFT-1 has better hypolipidemic effects compared with the ordinary tea fuyun6,and its mechanism may be related to improve the antioxidant activity,anti-inflammatory effects and regulate fat metabolism.
